Overview

CJC-1295 DAC is a long-acting synthetic analog of growth hormone-releasing hormone (GHRH) conjugated with a Drug Affinity Complex (DAC). This unique modification dramatically extends the peptide's half-life through reversible binding to serum albumin, making it one of the longest-acting peptide research compounds available.

The DAC technology was developed to overcome the inherently short half-life of native GHRH and its analogs. By attaching a reactive group that covalently binds to albumin's Cys34 residue in the bloodstream, the conjugate achieves a circulating half-life of approximately 8 days, compared to ~30 minutes for the unconjugated CJC-1295 (no DAC).

This extended duration of action results in sustained, continuous GH elevation rather than pulsatile release, making CJC-1295 DAC a distinct research tool for studying the effects of chronic versus pulsatile GH axis stimulation.

CJC-1295 DAC shares the same base GHRH receptor agonist mechanism as CJC-1295 (no DAC), activating the Gs/adenylyl cyclase/cAMP/PKA signaling cascade in pituitary somatotroph cells to promote GH synthesis and release.

The DAC component is a maleimidopropionic acid linker that, once injected, spontaneously forms a covalent bond with the free thiol group of Cys34 on serum albumin. This albumin conjugation serves multiple purposes: it protects the peptide from proteolytic degradation, reduces renal clearance (albumin is too large for glomerular filtration), and creates a circulating reservoir that slowly releases active peptide.

The resulting pharmacokinetic profile produces sustained rather than pulsatile GH elevation, with measurable increases in both GH and IGF-1 lasting for days after a single administration.

Handling & Reconstitution

1. 1Allow the vial and sterile water to reach room temperature (15–25°C).
2. 2Using a sterile syringe, withdraw the appropriate volume of sterile water.
3. 3Inject the water slowly along the inside wall of the vial — do not spray directly onto the powder.
4. 4Gently swirl the vial until the powder is fully dissolved. Do not shake vigorously.
5. 5Once reconstituted, store at 2°C – 8°C. Protect from light.
6. 6Use within 28 days after reconstitution.
