Semax

Overview

Semax is a synthetic heptapeptide derived from the ACTH(4-10) fragment with an additional Pro-Gly-Pro (PGP) tripeptide extension at the C-terminus. Developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, Semax represents one of the most extensively studied nootropic peptides.

The PGP extension serves dual purposes: it enhances the peptide's stability against enzymatic degradation and contributes additional biological activity through its own immunomodulatory properties. Semax has been the subject of extensive research in Russia, where studies have explored its effects on cognitive function, neuroprotection, and neuroplasticity.

Mechanism of Action

Semax's primary mechanism involves upregulation of brain-derived neurotrophic factor (BDNF) and its receptor tropomyosin receptor kinase B (TrkB). The BDNF/TrkB signaling cascade promotes neuronal survival, synaptic plasticity, and neurogenesis — processes fundamental to learning, memory, and cognitive function.

Additionally, Semax modulates serotonergic and dopaminergic neurotransmitter systems. It influences the expression and activity of tryptophan hydroxylase and tyrosine hydroxylase, the rate-limiting enzymes in serotonin and dopamine synthesis respectively. This dual modulation may contribute to both cognitive enhancement and mood-regulating effects observed in preclinical studies.

The PGP extension activates the proteolytic enzyme prolyl endopeptidase, generating additional bioactive fragments that may contribute to immunomodulatory and anti-inflammatory effects in the CNS.

Handling & Reconstitution

1. 1Allow the vial and sterile water to reach room temperature (15–25°C).
2. 2Using a sterile syringe, withdraw the appropriate volume of sterile water.
3. 3Inject the water slowly along the inside wall of the vial — do not spray directly onto the powder.
4. 4Gently swirl the vial until the powder is fully dissolved. Do not shake vigorously.
5. 5Once reconstituted, store at 2°C – 8°C. Protect from light.
6. 6Use within 28 days after reconstitution.
7. 7Do not freeze reconstituted solution.
