Thymosin Alpha-1

Overview

Thymosin Alpha-1 (Thymalfasin) is a 28-amino acid endogenous peptide originally isolated from the thymus gland. First characterized by Dr. Allan Goldstein in the 1970s, it represents the major biologically active component of Thymosin Fraction 5, a partially purified thymic extract that demonstrated immune-reconstituting properties.

Thymosin Alpha-1 plays a crucial role in T-cell development and maturation within the thymus. As a naturally occurring immunomodulator, it has been extensively studied for its ability to restore and enhance immune function, particularly in immunocompromised states.

Mechanism of Action

Thymosin Alpha-1 exerts its immunomodulatory effects primarily through activation of toll-like receptors (TLR2 and TLR9) on dendritic cells and other antigen-presenting cells. This TLR activation triggers MyD88-dependent signaling cascades that enhance the maturation and antigen-presenting capacity of dendritic cells.

The peptide promotes the differentiation and maturation of T-cell precursors into functional CD4+ helper and CD8+ cytotoxic T lymphocytes within the thymus. It enhances NK cell cytotoxicity by upregulating activating receptors (NKG2D, NKp46) and stimulates the production of key cytokines including IL-2, IFN-α, and IFN-γ.

Additionally, Thymosin Alpha-1 has been shown to modulate regulatory T-cell (Treg) function, helping to balance immune activation with tolerance — a property that may explain its observed benefits in autoimmune and inflammatory contexts.

Handling & Reconstitution

1. 1Allow the vial and sterile water to reach room temperature (15–25°C).
2. 2Using a sterile syringe, withdraw the appropriate volume of sterile water.
3. 3Inject the water slowly along the inside wall of the vial — do not spray directly onto the powder.
4. 4Gently swirl the vial until the powder is fully dissolved. Do not shake vigorously.
5. 5Once reconstituted, store at 2°C – 8°C. Protect from light.
6. 6Use within 28 days after reconstitution.
7. 7Do not freeze reconstituted solution.
